Risulta primo nome in 10 lavori in extenso e Corresponding Author in 3. - L’attività di ricerca è rivolta ad indagare i meccanismi nervosi centrali coinvolti nella ritmogenesi respiratoria e nella mediazione di riflessi delle vie aeree, cercando anche di chiarire il ruolo dei neurotrasmettitori coinvolti. Recentemente, i meccanismi nervosi alla base della genesi del ritmo respiratorio sono stati investigati nel preparato “tronco dell’encefalo in vitro” di lampreda, un vertebrato con un sistema nervoso centrale relativamente semplice. Questo preparato presenta numerosi vantaggi per lo studio dei meccanismi neurochimici endogeni responsabili della genesi di un pattern motorio spontaneo; inoltre, esso contribuisce a fornire suggerimenti per ulteriori studi sui meccanismi di base che operano nelle reti neuronali respiratorie di vertebrati superiori, inclusi i mammiferi. L’attività di ricerca è stata rivolta essenzialmente al controllo nervoso della respirazione. Gli argomenti di ricerca sono stati i seguenti: 1. Ruolo di alcune aree bulbo-pontine e delle afferenze inibitorie laringee sul controllo nervoso della respirazione;
2. Ritmogenesi respiratoria nel coniglio;
3. Ritmogenesi respiratoria nella lampreda;
4. Studio dei meccanismi coinvolti nella genesi del riflesso della tosse;
5. Studio nell’uomo sia in condizioni fisiologiche che patologiche dei meccanismi alla base della tosse riflessa e volontaria.

Research activity
My research activity has been devoted to study central mechanisms involved in the respiratory rhythmogenesis and in the mediation and regulation of airway defensive reflexes, especially the cough reflex, in in vivo preparations of rabbits. Mechanisms underlying respiratory rhythmogenesis have been also investigated using the in vitro brainstem-spinal cord preparation of the lamprey, a vertebrate with a simple CNS. This preparation offers several advantages for studying the endogenous neurochemical mechanisms underlying the production of a spontaneously generated motor pattern and contributes to get further insights into the neural mechanisms operating in the respiratory network of higher vertebrates, including mammals. Integrated approach combining electrophysiological, immunohistochemical and microinjection techniques in different animal models both in in vitro and in vivo preparations have been used.
Scientific achievements
- Control of breathing in in vivo preparations
Studies on the respiratory role played by different bulbar and pontine structures involved in the respiratory rhythmogenesis by using microinjections of different neuroactive agents in anaesthetized, vagotomized, paralysed and artificially ventilated rabbits. The results obtained in several studies demonstrate the complexity of the bulbar respiratory network and suggest that the rostral regions of the ventral respiratory group (VRG), i.e. the Bötzinger complex and the pre-Bötzinger complex, are important components of the neural network responsible for the generation of the eupneic pattern of breathing, thus supporting the view that the respiratory CPG is a distributed neural system characterized by a great deal of redundancy.
- The cough reflex in in vivo preparations
Studies on the bulbar neural structures involved in the mediation of airway defensive reflexes (cough and sneeze) in anaesthetized, spontaneously breathing rabbits to clarify the role played by different neurotransmitters and the action of antitussive drugs. It has been demonstrated that the blockade of glutamatergic neurotransmission within the nucleus tractus solitarii (NTS), the predominant site of central projections of tracheobronchial rapidly adapting receptors mainly involved in cough mediation, abolished or strongly reduced the cough reflex. It has been clarified that the caudal aspect of the NTS and the VRG may be a site of action of some centrally acting drugs (baclofen, DAMGO, NK1 receptor antagonists, cholinergic agents) and a likely component of a neural system involved in cough regulation. Evidence has been provided that local inhibition of ERK1/2 activation by microinjections of a MEK inhibitor into the nucleus tractus solitarii decreases or completely abolishes cough responses. Studies on this matter may provide fruitful strategies for the development of novel antitussive therapies.
- Control of breathing in in vitro preparations
Studies on nervous mechanisms involved in the genesis of the respiratory rhythm investigated using the "in vitro brainstem-spinal cord preparation" of the lamprey, a lower vertebrate with a simple CNS. Several physiological and pharmacological characteristics are retained throughout evolution. In these studies on the lamprey respiratory network, the role of glutamatergic transmission mediated by mGluRs, of the GABAergic and glycinergic transmission, of the opioids and μ, δ and κ receptors, of the neurokinins, some ionic currents, the cholinergic and serotoninergic transmission has been investigated. A region, called the paratrigeminal respiratory group (pTRG), has been characterized by using extracellular and intracellular neuronal recordings as well as pharmacological activation of receptors for different neurotransmitters. This region likely plays a pivotal role in the lamprey respiratory rhythmogenesis and has some characteristics similar to the mammalian pre-Bötzinger complex, a structure important for respiratory rhythmogenesis.
