Massimo Pinzani presso l'Istituto di Medicina Interna dell'Università di Firenze diretto dal Prof. Paolo Gentilini. Conseguimento del titolo di Dottore di Ricerca in data 8/1/1996 con la seguente tesi : “Espressione e regolazione delle binding proteins per gli insulin-like growth factors in cellule stellate perisinusoidali isolate da fegato umano”.Research Fellowship : Dal novembre 1994 al maggio 1996 presso il Dipartimento di Nefrologia dell'Università del Texas Health Science Center diretto dal Prof. Hanna E. Abboud.Postdottorato di Ricerca in Fisiopatologia Clinica: Iniziato nell'agosto 1997 presso l'Istituto di Medicina Interna dell'Università di Firenze, è terminato nell'agosto 1999. Argomento della Ricerca : “Rapporti biologici esistenti tra cellule epatocarcinomatose e la matrice extracellulare che costituisce lo stroma proprio del tumore”.Assegno di Ricerca : Iniziato nel settembre 1999 con il seguente progetto : “Meccanismi di segnalazione intracellulare dell’insulin-like growth factor I in cellule stellate umane” e terminato nel settembre 2000.Contratto triennale come coordinatore tecnico, categoria EP1: Iniziato il primo settembre 2000 e terminato nell’ottobre 2002. Assunzione come ricercatore a tempo indeterminato: dal 1° novembre 2002.Assunzione come ricercatore di ruolo : dal 1° novembre 2005.
ATTIVITA' DI RICERCA SCIENTIFICA
La Dott.ssa Gentilini ha frequentato dal 1989 al 1991 l'Istituto di Biochimica di Firenze dove ha svolto attività di ricerca presso i laboratori del Prof. A. Berti e Prof. G. Cappugi. Durante questo periodo si e' occupata prevalentemente di un progetto riguardante la purificazione e caratterizzazione dell'enzima acilfosfatasi da muscolo scheletrico di tacchino che e' stato il tema riguardante la sua tesi di laurea.Durante il Dottorato di Ricerca in Fisiopatologia Clinica ha contribuito a vari progetti nell'ambito dello studio della fisiopatologia delle Cellule Stellate Perisinusoidali (CSP) e della patogenesi della fibrosi epatica. In particolare i suoi studi si sono focalizzati su: 1) Regolazione dell'espressione delle subunita' recettoriali per il platelet-derived growth factor (PDGF), da parte del transforming growth factor-beta 1 (TGF-ß1), in CSP umane.2) Espressione e ruolo fisiopatologico-prognostico dell'espressione del platelet-derived growth factor e delle relative subunità recettoriali in corso di attive fibrogenesi epatica.3) Ruolo dell’attivazione della phosphatidylinositol 3- (PI 3- ) kinase nella mitogenesi e chemotassi indotte dal platelet-derived growth factor in CSP umane.4) Regolazione dell'espressione del gene per il procollagene tipo 1 da parte di meccanismi lipoperossidativi e di aldeidi reattive in CSP umane.5) Espressione di endotelina-1 (ET-1) nel fegato umano e cirrotico ed effetti biologici dell'ET-1 in CSP umane.6) Caratterizzazione e regolazione delle insulin-like growth factor binding proteins (IGFBPs) nelle CSP umane.Ha iniziato presso il Department of Medicine, University of Texas at San Antonio, Texas, U.S.A., sotto la diretta supervisione della Dott.ssa Sherry Abboud , un progetto riguardante l’espressione di IGF-I e IGFBPs nel fegato umano normale, cirrotico e neoplastico che è stato terminato in seguito presso il laboratorio di biologia molecolare e cellulare di Medicina Interna di Firenze coordinato dal Prof. M.Pinzani. Svolge tuttora attività di ricerca nel campo del signaling dell’IGF I presso lo stesso laboratorio, soprattutto nell’ambito delle attività di fattore di crescita e di sopravvivenza cellulare svolte da tale citochina.
TECNOLOGIE DI BIOLOGIA MOLECOLARE ACQUISITE
• Purificazione di anticorpi monoclonali e policlonali.• Purificazione di proteine tramite precipitazione con solfato d'ammonio, centrifugazione frazionata, cromatografia a scambio ionico ed esclusione, ed HPLC.• Immunoprecipitazione di proteine ottenute da lisati cellulari.• Quantificazione e separazione su gel di poliacrilamide di proteine cellulari, con successiva identificazione o mediante anticorpi mono o policlonali secondo la metodica di western blot o con ligandi radioattivi secondo la metodica di ligand blot.• Coltivazione di cellule in vitro.• Tecnologie generali di DNA ricombinante: crescita di vettori plasmidici e purificazione di sonde di DNA, ligazione di sonde a vettori diversi e successivo subcloning. Marcatura di tali sonde con fosforo radioattivo.• Purificazione di RNA totale da cellule in coltura e da frammenti di tessuto epatico e successiva separazione su gel di agarosio/formaldeide (metodica Northern) o di urea/poliacrilamide (metodica RNase protection assay). Purificazione di sonde di RNA e marcature con fosforo radioattivo.• Metodologie per lo studio della proliferazione cellulare (incorporazione di timidina triziata) e della chemotassi (migrazione in camere di Boyden modificate).• Metodologie per lo studio della signal transduction, in particolare per lo studio dell'attivazione della mitogen activated protein (MAP) kinase, della PI3 kinase e della protein kinase B.• Metodologie per lo studio dell’apoptosi, in particolare l’espressione dell’attività enzimatica per la Caspase-3 e la regolazione di proteine impiegate nel signaling, quali Bcl-xl e Bad.

RESEARCH ACTIVITIES
As preparation for the Biological Science Degree thesis, purification and characterization of an acylphosphatase from turkey skeletal muscle.
For Ph.D. program partecipation in various projects directed mainly by M. Pinzani M.D. Ph.D., concerning the role of the Human Hepatic cells (HSCs) in the development of liver fibrosis.
These projects included :
-Effects of PDGF and/or TGF- on HFSC DNA synthesis and TGF- regulation of PDGF receptor expression.
-Role of phosphatidylinositol 3 (PI3)-kinase activation in PDGF induced mitogenesis and chemotaxis in HFSC.
-Regulation of expression of procollagen I mRNA by lipoperoxidative mechanisms in HFSC.
-Endothelin 1 expression in normal and cirrhotic livers and mitogenic effects of Endothelin 1 in HFSC.
As Research Fellow in San Antonio, under the supervision of Sherry Abboud M.D. Ph.D., research activity in a project studying the characterization and regulation of Insulin-like growth factor (IGF)-binding proteins in HFSC. Partecipation in two projects with G.Ghosh Choudhury, Ph.D. studying the involvement of mitogen activated protein (MAP) kinase in glomerular mesangial cells on PI3-kinase dependent chemotaxis and mitogenesis and the involvement of protein kinase C.
Back in Florence research activity in a project studying the profibrogenic action of IGF-I on HSC. Partecipation as collaborator or principal investigator in different projects studying i) the interactions between stromal cells and cholangiocarcinoma and ii) liver fibrosis.
MOLECULAR AND CELL BIOLOGY TECHNIQUES
• Purification of monoclonal and polyclonal antibodies.
• Protein purification by ammonium sulfate precipitation, fractional centrifugation, ion exchange and exclusion chromatography, and HPLC.
• Immunoprecipitation of proteins obtained from cellular lysates.
• Quantification and separation of cellular proteins on polyacrylamide gel, with subsequent identification either by mono or polyclonal antibodies according to the western blot method or with radioactive ligands according to the ligand blot method.
• In vitro cell culture.
• Technologies of recombinant DNA: growth of plasmid vectors and purification of DNA probes, ligation of probes to different vectors and subsequent subcloning. Labeling of probes with radioactive phosphorus.
• Purification of total RNA from cultured cells and liver tissue fragments and subsequent separation on agarose / formaldehyde gel (Northern method) or urea / polyacrylamide (RNase protection assay method). Purification of RNA probes and radioactive labeling.
• Methods for the study of cell proliferation (incorporation of tritiated thymidine) and motility (migration an invasion with modified Boyden chambers).
• Methodologies for the study of signal transduction, in particular for the study of the activation of mitogen activated protein (MAP) kinase, PI3 kinase and protein kinase B.
• Methods for genes silencing by siRNA and shRNA.
